摘要
目的观察氨氯地平对氧化型低密度脂蛋白(ox-LDL)诱导的人脐静脉内皮细胞(hUVEC-12)单核细胞趋化蛋白1(MCP-1)mRNA表达的影响,以进一步探讨氨氯地平抗动脉粥样硬化的作用机制。方法筛选ox-LDL与hUVEC-12细胞孵育合适的处理浓度与时间,然后用不同浓度氨氯地平(0、0.1、1.0、10.0μmol/L)预孵育hUVEC-12后,与ox-LDL共同孵育,RT-PCR检测细胞MCP-1 mRNA表达的变化情况。结果筛选出ox-LDL与hUVEC-12共同孵育的合适浓度为50 mg/L,时间为24 h。氨氯地平可下调MCP-1 mR-NA的表达,并且对其表达的影响呈现浓度依赖性。结论氨氯地平可下调ox-LDL诱导的人脐静脉内皮细胞MCP-1 mRNA的表达。
Objective To investigate the effects of amlodipine on the mRNA expression of monocytes chemotactic protein - 1 { MCP - 1 ) in human umbilical vein endothelial cell ( hUVEC ) by oxidized low density lipoprotein ( ox - LDL ), so as to approach the mechanism of amlodipine in atherosclerosis. Methods hUVEC - 12 were treated with ox - LDL in different concentration or different time. Based on that, the cells were pretreated with amlodipine at different concentrations ( 0,0.1,1.0,10.0 μmol/L) and then incubated with ox - LDL, MCP - 1 mRNA expression was detected by reverse tran- scription - polymerase chain reaction ( RT - PCR). Results MCP - 1 mB_NA expression induced by ox - LDL was obviously down - regulated by aralodipine in a dose - dependent manner. Conclusion Amlodipine may down - regulate the mRNA expression of MCP - 1 induced by ox - LDL in hUVEC - 12.
