MIF抑制剂HA对胃癌细胞株BGC-823增殖和凋亡的影响

摘要 目的体外观察透明质酸(HA)抑制巨噬细胞移动抑制因子(MIF)后对人胃癌细胞株BGC-823增殖与凋亡的影响。方法以体外培养的人胃癌细胞株BGC-823为研究对象。采用MTT比色法研究HA抑制MIF后对BGC-823细胞增殖的影响;流式细胞术和AO—EB荧光染色研究HA抑制MIF后对BGC-823细胞凋亡的影响;流式细胞术分析HA抑制MIF后对BGC-823细胞周期的影响。结果MTT试验显示100-400μg/mL HA抑制MIF后对BGC-823细胞增殖具有抑制作用,呈浓度和时间依赖性。AO—EB染色和流式细胞术显示BGC-823细胞的凋亡率、破膜率随HA浓度的升高对MIF抑制作用的增强而增加。流式细胞术检测发现,HA作用细胞48h后能将细胞阻滞于G0/G1期,呈浓度依赖性。结论MIF促进人胃癌BGC-823细胞增殖,抑制其凋亡,HA可抑制其作用。 Objective To observe the effect of macrophage migration inhibitory factor of hyaluronic acid on proliferation and apoptosis in human gastric cancer cell line BGC - 823 in vitro. Methods MTT assay was used to determine the effect on the inhibition of the growth of BGC - 823 cell by the drugs. Flow cytometry and AO - EB fluorescent staining were used to determine the effect on the apoptosis of BGC - 823 cells by the drugs. Flow eytometry was used to assay the effect on the cell cycle of BGC - 823 ceils by the drugs. Results The result of MTT assay showed that the growth of BGC - 823 cells was inhibited by HA inhibition to MIF with concentration from 100 μg/mL to 400 μg/mL, and the effect increased in a concentration - and time - dependent manner. The results of apoptosis on the cell cycle of BGC - 823 cells determined by AO - EB fluorescent stainining and flow cytometry were coincident: compared with the negative control group, apoptosis of HA group was significantly increased, and the rates of apoptosis and rupture of membrane were increased with higher con- centration. The result of flow cytometry showed that HA could block off BGC - 823 ceils in stage of G0/G1, and the role was significantly increased with higher concentration compared with the negative control group. Conclusions MIF can promote the growth and inhibit the apoptosis of human gastric cancer ceils BGC - 823.
