肿瘤坏死因子-α在大鼠急性脊髓损伤中的表达 被引量:9

摘要 目的探讨肿瘤坏死因子(TNF)-α与大鼠急性脊髓损伤的关系。方法按改良Allen′s撞击法制作脊髓损伤模型,用ELISA方法检测大鼠脊髓损伤0、24、48、72、96hTNF-α的表达,并用RT-PCR法测定上述大鼠损伤组织中TNF-αmRNA表达量。结果ELISA法检测结果显示,在脊髓损伤24h后,TNF-α表达明显增加,与0h组比较有显著性差异(P<0.05)。与24h组比较,48、72h及96hTNF-α表达进一步明显增加(P<0.05)。RT-PCR结果显示,在脊髓损伤24h后,TNF-αmRNA表达明显增加,与0h组比较有显著性差异(P<0.05);随着损伤时间的延长,TNF-αmRNA在损伤后24、48、72h及96h表达并无明显差异。结论TNF-α可能是加重脊髓损伤的一个重要因素,抑制TNF-α的表达可能会成为治疗脊髓损伤的一个有用手段。
