摘要
目的:观察青少年特发性脊柱侧凸(adolescent idiopathic scoliosis,AIS)患者软骨细胞中褪黑素受体含量的变化,并探讨其与AIS病因学的关系。方法:随机选取2007年1月-2007年12月在我院手术治疗的22例AIS患者及8例非脊柱侧凸(肿瘤、外伤等)患者(对照组)。AIS组患者年龄10-16岁,平均12.7岁,平均Cobb角60.1°,其中10例在术中取髂软骨,12例取棘突软骨。对照组患者年龄10-15岁,平均12.9岁,其中7例在术中取髂软骨,1例取棘突软骨。对髂软骨和棘突软骨标本进行细胞培养至二代,取二代细胞用Ⅱ型胶原免疫组化染色方法进行软骨细胞表型鉴定,并抽提总RNA,采用逆转录聚合酶链反应(RT-PCR)检测软骨细胞中褪黑素受体两种亚型MTNR1A、MTNR1B的mRNA表达量。结果:两组患者软骨细胞Ⅱ型胶原免疫组化染色均为阳性表达。AIS组软骨细胞中MTNR1A、MTNR1B mRNA的相对表达量分别为0.44±0.29和0.54±0.31.对照组分别为0.69±0.18和0.83±0.30,组间比较均有显著性差异(P〈0.05)。结论:AIS患者软骨细胞中MTNR1A、MTNR1B的mRNA表达均减少,这可能与AIS的发生、发展有关。
Objective:To investigate the expression of melatonin receptors in chondrocyte derived from adolescent idiopathic scoliosis (AIS) patients,and explore the role of melatonin in the pathogenesis of seoliosis. Method:From January 2005 to December 2005 ,two groups were defined as AIS group and control group.AIS group consisted of 22 patients with an average age of 12.7 years,ranging from 10 to 16 years.The average Cobb angle was 60.1°.10 patients' iliac cartilage and 12 patients' spinal process cartilage were taken during spinal surgery.Control group consisted of 8 subjects with an average age of 12.9 years,ranging from 10 to 15 years who underwent surgery because of benign tumors,trauma.7 patients' iliac cartilage and 1 patients' spinal process cartilage were taken during surgery.The chondrocyte from iliac cartilage and spinal process cartilage were cultivated and serial subcultivated in vitro.Immunohistocbemical stain of collagen Ⅱ were performed to confirm the P2 chondrocyte phenotype.The mRNA expression of mclatonin recepter IA(MTNR1A) and melatonin receptor 1B(MTNR1B) in chondrocyte were detected by RT-PCR method.Result:The collagen type 11 immunohistochemical staining was positive in cells of two groups.The mRNA expression level of MTNR1A and MTNR1B was 0.44±0.29 and 0.54±0.31 in AIS group and 0.69±0.18 and 0.83±0.30 in control group respec-tively,which showed significant difference (P〈0.05).Conclusion:Decreased expression of MTNR1A and MT- NR1B in chondrocyte of AIS patients were detected.This abnormal expression of melatonin receptor in AIS patients show that melatonin may be related to the pathogenesis of AIS.
