摘要
目的观察骨缺损区随时间变化骨断端组织成分和核心蛋白多糖(decorin,DCN)含量的变化,分析两者的关系。方法选取新西兰大白兔,于桡骨中段截除1.0 cm骨段,去除骨外膜,石蜡封闭髓腔,分别于术后4、6、8周后行大体观察、影像学和组织学检测,比较骨断端组织成分的变化,免疫组化检测组织中DCN的表达。结果随着骨缺损形成时间延长,骨断端的骨痂逐渐减少,组织成分渐向纤维瘢痕组织变化,DCN的表达在骨缺损形成8周时明显降低。结论骨缺损区组织纤维瘢痕化转变与局部组织成骨能力低下和DCN分泌减少密切相关。
Objective To Observe the changes of tissue composition and decorin content of bone defects over time and analyze the relationship between them. Methods New Zealand rabbits were selected, in the middle of the radial bone 1.0 cm bone was cut, periosteum was removed, paraffin cavum medullare was closed by paraffin macroscopic observation, radiological and histological detection, omparison changes of tissue composition were examined, the expression of DCN in the tissue was examined by immunohistochemical 4 ,6,8 weeks later. Results With the time of the bone defect formation, the bone callus became diminished gradually , bone tissue composition transformed to fiber scar tissue, the expression of DCN at eight weeks decreased most significantly. Conclusion Local tissue transforming to scar tissue fibers and the low osteogenesis capacity at site of bone defect related to the reduction of the DCN secretion.
