摘要
目的设计用于专门进行肿瘤内睾丸特定基因—癌-睾丸(cancer-testis,CT)基因转录分析的微阵列的探针系列.以阐明CT基因在肿瘤发生过程中的作用并检测它们在肿瘤细胞或肿瘤样本中的表达。方法通过二个探针设计工具,IDT和MEDIANTE的结合使用,设计出CT基因的寡核苷酸探针。同时,应用“Oligonucleotide Properties Calculator寡核苷酸性质计算软件”和“OligoAnalyzer 3.0”检测探针的解链温度(Tm)、GC含量和可能形成的次级结构等特性。结果为CT基因家族的44个成员成功设计了113个50mers左右的寡核苷酸探针。其Tm值分布在63.2℃~67.5℃之间,GC百分含量为35.3%~51.5%。结论结合IDT和MEDIANTE两个探针设计软件,能有效的进行CT基因寡核苷酸探针的设计。
Objective To design oligonucleotide probes for Cancer-testis (CT) genes for a microarray, which will be dedicated to a transcriptomic analysis of testis-specific genes in tumors and elucidation of the role of CT genes in the process of tumorigenesis. Methods The oligonucleotide probes for CT genes were designed through a combination of two probe designing tools available online, IDT and MEDIANTE. The "Oligonucleotide Properties Calculator" and "OligoAnalyzer 3.0" were applied to check the probe properties, including melting temperature, GC content and secondary structure forming possibilities etc. Results A total of 113 of 50 mer oligonueleotide probes were successfully designed for the 44 families of CT genes. The probes had a narrow range of Tm values between 63.2 ℃ and 67.5 ℃ and GC% contents between 35.3% and 51.5%. Conclusion The combinative use of the two probe designing tools, IDT and MEDIANTE, can successfully design oligonucleotide probes for cancer-testis genes.
