食管癌组织STRBP8基因克隆及其生物信息学分析

The gene cloning and bioinformatic analysis of STRBP8 from esophageal cancer tissues

目的:克隆食管癌组织核基质蛋白类视网膜母细胞瘤结合蛋白8(STRBP8)CDS编码序列,为建立其免疫学检测技术奠定基础。方法:从GenBank中获取STRBP8的基因序列,并根据其CDS区序列设计全长引物。利用Trizol法从食管癌组织中提取总RNA,通过逆转录获取STRBP8的cDNA,再利用梯度升温PCR方法对目标基因进行扩增。结果:本实验成功从食管癌组织中克隆出STRBP8的CDS序列。结论:STRBP8在食管癌组织均存在表达,为下一步建立其免疫学检测技术打下基础。

Objective ： To clone the CDS coding sequence of similar to retinoblastoma binding protein 8 （STRBP8） from esophageal cancer tissues, and get prepare for establishment of immunological detection technology. Methods： The corresponding primers were designed according to the STRBP8 sequence from GenBank. The target gene of cDNA was amplified by Step-up PCR. Results.. The CDS sequence of STRBP8 was cloned successfully from the sophageal cancer tissues. Conclusions： STRBP8 is expressed in esophageal cancer tissues, which lays foundation for further studies on the establishment of immunological detection technology .
