摘要
目的:克隆食管癌组织核基质蛋白类视网膜母细胞瘤结合蛋白8(STRBP8)CDS编码序列,为建立其免疫学检测技术奠定基础。方法:从GenBank中获取STRBP8的基因序列,并根据其CDS区序列设计全长引物。利用Trizol法从食管癌组织中提取总RNA,通过逆转录获取STRBP8的cDNA,再利用梯度升温PCR方法对目标基因进行扩增。结果:本实验成功从食管癌组织中克隆出STRBP8的CDS序列。结论:STRBP8在食管癌组织均存在表达,为下一步建立其免疫学检测技术打下基础。
Objective : To clone the CDS coding sequence of similar to retinoblastoma binding protein 8 (STRBP8) from esophageal cancer tissues, and get prepare for establishment of immunological detection technology. Methods: The corresponding primers were designed according to the STRBP8 sequence from GenBank. The target gene of cDNA was amplified by Step-up PCR. Results.. The CDS sequence of STRBP8 was cloned successfully from the sophageal cancer tissues. Conclusions: STRBP8 is expressed in esophageal cancer tissues, which lays foundation for further studies on the establishment of immunological detection technology .
