摘要
以氯乙酸和二氯亚砜为原料用传统的方法合成氯乙酰氯,进而以其为酰化试剂,以对氨基酚为原料,在氯乙烷和乙酸乙酯溶剂中通过正交实验分别确定了氯乙烷为最佳溶剂和该反应的最佳反应条件为:以氯乙烷为溶剂时,n(对氨基酚)∶n(氯乙酰氯)∶n(溶剂)=1∶2.5∶9,反应温度80℃,反应时间3 h,最高收率为90%。同时进行了规律性实验和强化实验证明了上述最佳反应条件。并对所合成的产物进行了熔点测定、元素分析、质谱分析、红外光谱分析以及核磁共振氢谱分析,确定所合成的化合物即是目的产物。
Chloroacetyl chloride was synthesized from monochloroacetic acid and thionyl dichoride by conventional method.Then,2-chloro-N-(4-hydroxyphenyl) acetamide was synthesized in a mixed solvent composed of chloroethane and ethyl acetate by the reaction of carbonyl chlorinating agent-chloroacetyl chloride with 4-aminophenol.The product yield was more than 90% in chloroethane solvent.Moreover,effects of reaction conditions on the product yield were discussed.The optimum preparation conditions were obtained by orthogonal experiments as follows:a ratio of n(4-aminophenol)∶n(chloroacetyl chloride)∶n(solvent)=1∶2.5∶9,a reaction temperature of 80 ℃,and a reaction time of 3 h.The structure of target product was verified by means of EA,mass spectrometry,IR and 1HNMR spectroscopy.
