摘要
以贵农1号、2号、5号、6号、7号刺梨为材料,研究不同基因型、不同取材时间、小孢子发育时期、基本培养基、糖源、生长调节物质、培养条件等刺梨花药愈伤组织诱导的影响。结果表明;以刺梨盛花期(5月份)且小孢子发育为单核中后期的贵农1号花药为试材;Ms为基本培养基;3%的蔗糖为糖源;1.0 mg.L-16-BA+2.0mg.L-1NAA+0.5 mg.L-12、4-D+1.0 mg.L-1KT为激素组合,在黑暗条件下培养最有利于刺梨花药愈伤组织的诱导。
To obtain anther callus of Rosa roxburghii Tratt, five genotypes including Guinong 1, Guinong 2, Guinong 5, Guinong 6, and Guinong 7 were employed to carry out various induction experiments in the present investigation. The resuits demonstrated that several factors, including genotype, developmental stage of microspore, basal medium, carbonic source, plant growth regulator ( PGR), as well as incubating condition may have diverse effects on the callus formation. Anther of Guinong 1 obtained at full - blossom stage, whose development is around mid - anaphase of uninucleate microspore, was proven to be the best for the induction. Murashige and Skoog (MS) basal medium, supplemented with the combination of1.0 mg.L-16-BA, 2.0 mg.L-1 NAA, 0.5 mg.L-1 2,4-D, 1.0 mg. L-1 KT, and3% sucrose gave the optimal response. Additionally, the cultures incubated in dark condition somewhat showed improved effect on callus induction.
