从西双版纳红木种子中制备胭脂树橙的工艺研究

摘要 对西双版纳红木种子中胭脂树橙的含量、提取、晶体制备的工艺条件以及所制备产品的色价、纯度、结构等进行了研究。结果表明:西双版纳红木种子中胭脂树橙含量为1.2%;百克级提取工艺中选择乙酸乙酯作提取溶剂,料溶比1∶3,温度60℃,时间20 min,搅拌提取3次,提取率为(85±1)%;结晶纯化工艺用乙醇作结晶溶剂,在5℃环境中静置3.5 h,可得到UV含量98.3%,色价282.7的流动性胭脂树橙结晶性粉末,结晶收率90%,重结晶收率83.3%;通过质谱MS、核磁共振谱(1H-NMR和13C-NMR)表征了胭脂树橙的化学结构,表明所确定的工艺条件制备的产品纯度较高且未同分异构化和热降解。 The content, color scale, purity and structure of bixin in annatto seeds in xishuangbanna and its technology conditions of extracting and crystal preparation were studied e in this paper. Results showed that the content of bixin in annatto seeds in xishuangbanna was 1.2%. Extracting conditions of 100 -g level were as follows: ethyl acetate was the solvent, the ratio of raw material to solvent was 1 : 3, the temperature was 60 ℃, the time was 20 min and extracting for 3 times. On this condition, the extracting rate of bixin was (85 ± 1 ) %. The condition of optimum purification was standing with ethanol solution at 5℃ for 3.5 h, and the crystal of bixin was obtained, its content was 98.3 %, color scale was 282. 7, the crystal rate was 90% and the recrystal rate was 83.3%. The chemical composition and structure of the bixin were characterized by MS, ^1H-NMR and ^13C-NMR, which showed it had higher purity and did not produce trans-cisisomerization and thermal degradation.
