摘要
目的对比骨髓干细胞移植、皮下注射重组人粒细胞集落刺激因子(recombinant human granulocytecolonystimulating factor,rhG-CSF)、骨髓干细胞移植联合 rhG-CSF 皮下注射3种方法对糖尿病兔缺血组织血管新生的作用效果。方法健康新西兰兔,用四氧嘧啶(alloxanate,ALX)诱导糖尿病后,结扎兔右后肢股动脉,建立糖尿病下肢缺血动物模型,随机分成4组,A 组(rhG-CSF 动员组),B 组(rhG-CSF 动员+局部干细胞移植组),C 组(局部干细胞移植组),D 组(PBS 对照组)。治疗28 d 后全麻,4组均以右侧股内侧正中部位为中心等距离取局部肌肉组织,制作切片,免疫组织化学染色,检测各组骨骼肌微血管密度及血管内皮细胞生长因子(vascular endothelialgrowth factor,VEGF)表达水平。结果①A、B、C 组均使局部缺血组织微血管数量增加,B 组微血管数量明显高于其他2组(P<0.05)。②A、B、C 3组局部缺血组织 VEGF 的表达量均增加,但 B 组 VEGF 的表达量明显高于其他2组(P<0.05)。结论 rhG-CSF 动员后联合骨髓干细胞移植治疗糖尿病下肢缺血性疾病,其效果明显优于单纯用 rhG-CSF 治疗和单纯骨髓干细胞移植治疗。
Objective To contrast the effect on neovascularization of the three methods, transplantation;Group D, control group. Twenty-eight days later, rabbits of four groups were executed. Partial musculatures were stained by the method of immunohistochemistry to detect the expression of yon willebrand factor (vWF) and vascular endothelial growth factor (VEGF). Results ①The expression of vWF was increased in group A, B and C, while group B was more intense than group A and C. ②The expression of VEGF was increased in group A,B and C,and it was the highest in group B. Conclusion The method of combining bone marrow stem cell transplantation with hypodermic injection granulocyte-colony stimulating factor (G-CSF) in treating diabetic lower limb ischemia was better than the treatment with single one.
