摘要
目的探讨 PTEN、P73蛋白表达与乳腺癌转移之间的关系。方法采用免疫组织化学链霉菌抗生物素-过氧化物酶法检测30例乳腺癌患者术后出现远处转移癌组织中 PTEN 和 P73蛋白的含量,同时取同期无远处转移30例患者作为对照。结果 PTEN 在远处转移组、局部淋巴结转移多于4枚组的高表达率均低于无远处转移组和局部淋巴结转移4枚及不足4枚组(P<0.05),且随着组织学分级的增高其高表达率反而逐渐降低(P<0.05)。P73在远处转移组和无远处转移组之间的高表达率差异无统计学意义(P>0.05);在局部淋巴结转移4枚及不足4枚的患者较转移多于4枚的患者呈现低表达(P<0.05);且在组织学分级Ⅰ、Ⅱ、Ⅲ级3组中的高表达率分别为41.7%、75.O%、76.9%,3组间差异有统计学意义(P<0.01)。结论 PTEN 基因的低表达与乳腺癌的局部和远处转移存在一定的正相关关系。
Objective Most of breast cancer patients died of distant metastasis. There is still not a method of predicting and early diagnosing this metastasis. This study was conducted to investigate the relation between the protein expressions of phosphatase and tensin homology deleted on chromosome ten(PTEN) and P73 and breast cancer metastasis. Methods PTEN and P73 protein expressions were examined by using immunohistochemical SP staining method in 30 cases of breast cancer patients metastasized after radical mastectomy, and 30 cases of no metastasis in the same time as the control group. Results The expressions of PTEN in the breast cancer tissue with distant metastasis and more than 4 local lymph node metastasis were lower than that of no distant metastasis and 4 or less than 4 loeal lymph node metastasis, and with the increase of histological stage PTEN expression was gradually decreased (P 〈 0.05). The expressions of P73 had no significant difference between distant metastasis group and no distant metastasis group ( P〉0.05) ; but P73 expression in the group of 4 or less than 4 lymph node metastasis was lower than that of the more than 4 lymph node metastasis group; and with the increase of histological stage, P73 expression was gradually increased ( P 〈 0.01 ). Conclusion The low expression of PTEN relates to local and distant metastasis of breast cancer.
