摘要
用磁珠富集法构建了长鳍吻鮈(AAAG)n富集文库,从中分离并鉴定得到13个微卫星位点。对三峡库区重庆江段的40尾长鳍吻鮈群体进行了遗传多样性的初步研究,其中6个位点呈现多态性(1个位点为中度多态,5个位点为高度多态),等位基因数目为4~11个,观测杂合度在0.20~0.85。这些多态性位点将为长鳍吻鮈及其近缘种的种群遗传研究提供有力的遗传学资料。
From a (AAAG)n enriched genomic library, we developed 13 microsatellites loci from Rhinogobio ventralis, using the fast isolation by AFLP of sequences containing repeats (FIASCO) protocol. In a test on a population of 40 individuals from Three Gorges Reservoir, Six primer pairs exhibited polymorphic with 4 to 11 alleles per locus. Observed heterozygosities ranged from 0.20 to 0.85. These loci should provide sufficient level of genetic diversity to evaluate the fine - scale population structure in the population of Rhinogobio ventralis and its congeneric species.
