摘要
以冬青苦丁茶树的幼嫩枝梢为材料进行组织培养,结果表明:外植体在MT+BA 1 mg/L+2,4-D 0.05 mg/L培养基培养,腋芽萌发最快,萌发率最高;随着外植体木质化程度的增加,褐变率降低,腋芽萌动时间缩短,萌动率提高;5、8、10月份取材的外植体,萌动率5月>8月>10月,腋芽萌动时间则相反;MT+BA 0.5 mg/L+NAA 0.2 mg/L培养基能使试管小苗茎段快速增殖;试管苗在1/2MT+NAA 0.2 mg/L+IBA 0.2 mg/L培养基上短时间培养后,蛭石上扦插发根,较容易移栽成活。
Using young shoots of ilex holly tree as materials in tissue culture, the regeneration of plants was studied. The results showed that explants cultured in the MT + BA 1 mg/L +2,4-D 0.05 mg/L culture medium, axillary bud germinated fast, germination rate was the highest. With the increase of explants lignification degree, the browning rate decreased, germinating axillary time was shorter, and germinating rate became higher. Germinating rate of explants derived in different month was as an order of May〉 August〉 October, but axillary germinating time was opposite. The stems of test-tube seedling grew rapidly in MT + BA 0.5 mg/L + NAA 0.2 mg/L medium. Test-tube seedling cultured in vitro 1/2 MT + NAA 0.2 mg/L + IBA 0.2 mg/L culture medium after a short time, rooting cuttings on vermiculite, transplanted survival easily.
