低强度He-Ne激光照射对离体小鼠巨噬细胞Ca^(2+)浓度的影响 被引量:3

摘要 为研究低强度He-Ne激光照射对离体小鼠巨噬细胞内Ca2+浓度的影响以及其与能量密度(剂量)的关系,用Fluo-3乙酰甲脂对巨噬细胞Ca2+作荧光标记,应用图象分析系统检测激光照射后细胞内Ca2+的荧光强度变化。结果:He-Ne激光照射引起细胞内Ca2+浓度改变。照射5min,激光剂量为0.679J/cm2,细胞荧光强度出现非常明显的增强;照射10min,剂量为1.358J/cm2,细胞荧光强度增强达到最大值;照射15min,剂量为2.037J/cm2,细胞荧光强度减弱;照射20min,剂量为2.716J/cm2,细胞荧光强度降低至对照组之下。低强度He-Ne激光照射巨噬细胞可引起细胞内Ca2+浓度上升,产生细胞内信号传递,进而控制细胞功能。 he purpose of this study is to investigate the relation of HeNe laser irradiation and its energy intensity and the intracellular calcium concentration in mouse macrophages. The intracellular calcium was labeled with Fluo3 AM and the fluorescence intensity of the macrophages after laser irradiatiion in vitro was determined by a VIDAS image analysis system. Changes of the intracellular Ca2+ concentrations were revealed after HeNe laser irradiation. The cellular fluorescence intensity increased obviously after 5 minutes irradiation with a light dose of 0679 J/cm2. The maximum value of the cellular fluorescence intensity was observed after 10 minutes laser irradiation with a light dose of 1358 J/cm2. A decreasement of the cellular fluorescence intensity occurred at 15 minutes irradiation with a light dose of 2037 J/cm2, and the intracellular fluorescence intensity was lower than that of control group after 20 minutes laser irradiation with a light dose of 2716J/cm2. The rise of the intracellular Ca2+ concentration in macrophages induced by low power HeNe laser irradiation may affect the intracellular signal transmission, controlling the cellular function. Our findings may provide certain evidence in explanation of the mechanisms of clinical HeNe laser therapy.
