系膜增生性肾炎肾组织E2F-1的表达及意义

摘要 目的探讨细胞周期相关核转录因子E2F-1在系膜增生性肾炎(MsPGN)发病机制中的作用。方法采用免疫组织化学SABC法检测石蜡包埋的36例MsPGN患者肾活检组织和6例外伤切除肾组织E2F-1的表达与分布,分析E2F-1与肾固有细胞增殖程度的关系。结果正常对照组肾小球未见E2F-1表达,肾小管间质见少许表达。MsPGN组肾小球和小管间质均有E2F-1表达,两组比较,差异具有显著性(P<0.01),各亚组随轻、中、重度系膜增生,E2F-1在肾小球和小管间质表达逐渐增强,亚组间比较差异有显著性(P<0.05或P<0.01)。结论E2F-1表达随系膜增殖程度加剧而增强,导致细胞周期调节紊乱,促进固有细胞增生,并可作为肾脏固有细胞增殖的标志。 [Objective] To investigate the role of nuclear transcription factor E2F-1 in renal tissue of patients with mesangial proliferative glomerulonephritis (MsPGN). [Methods] Paraffin-embedded renal biopsy renal tissue sections from 36 patiets with MsPGN were detected by immunohistochemical staining with anti-E2F-1 monoclone antibody. Normal renal tissue sections from 6 nephreetomized pateits with trauma were used as controls. Possible correlation between E2F-1 positive area and cell proliferation in renal tissue. [Results] There was no obvious expression of E2F-1 in nomal glomeruli and renal tubular-interstitial. Compared to the normal control group, it was significantly higher that the expression of E2F-1 in MsPGN group(P 〈0.01), the more severity of mesangial proliferation, the more expression of E2F-1 in glomeruli and renal tubular-interstitial. Furthermore, there was statistic difference among each subgroup (P 〈0.05 or P 〈0.01). [Conclusions] This study has demonstrated that the overexpression of E2F-1 in MsPGN. The more severity of proliferative, the more expression of E2F-1 in renal tissue, E2F- 1 promoted renal cell proliferation. So E2F-1 play important roles in mediating progressive renal damage in human MsPGN. In brief, it is a possibly important marker in renal proliferative cell of MsPGN.
