摘要
[目的]研究泥胡菜快速繁殖技术并建立高效无性系。[方法]以泥胡菜嫩茎为材料,研究愈伤组织诱导和分化、不定芽的分化和生根、试管苗移栽和扦插及移植于山坡栽培所需要的条件。[结果]MS+6-BA 0.3 mg/L+2,4-D 1.0-1.5 mg/L是诱导嫩茎形成颗粒状愈伤组织的理想培养基;MS+AgNO31.5 mg/L+6-BA 0.4 mg/L+NAA 0.1 mg/L是颗粒状愈伤组织和不定芽分化培养的理想培养基;1/2 MS+IAA 0.6 mg/L是试管苗生根和生根继代培养的理想培养基;炉灰渣是试管苗移栽和扦插的理想基质。[结论]该研究为泥胡菜组织培养中适宜的培养基选择提供科学依据。
[ Objective ] The research aimed to study the rapid propagation technology and establish effective clone of Hemistepta lyrata Bunge. [ Method ] With tender stem of Hemistepta lyrata Bunge as material, the conditions needed in callus induction and differentiation, adventitious bud differentiation and radication, test tube seedling cutting and transplantation were studied. [ Result] The results showed that the optimum medium for granulated callus indpction from tender stem was MS + 6-BA 0.3 mg/L + 2,4-D 1.0 - 1.5 mg/L, for granulated callus and adventitious bud differentiation was MS + AgNO3 1.5 mg/L + 6-BA 0.4 mg/L + NAA 0. 1 mg/L. 1/2 MS + IAA 0.6 mg/L was suitable for test tube seedling rooting and regeneration, and cinder was used as transplation and cutting substrate. [ Conclusion ] This study will provide the scientific reference for choosing the feasible medium in tissue culture of Hemistepta lyrata Bunge.
