摘要 [目的]研究牡荆总黄酮的最佳提取工艺及其抗氧化活性。[方法]以牡荆叶片为材料,采用单因素和正交试验研究乙醇浓度、固液比、超声时间及超声功率对总黄酮提取率的影响,在此基础上对提取工艺进行优化;以大鼠肝匀浆为材料,测定牡荆总黄酮的抗氧化活性。[结果]各因素对总黄酮提取率的影响由大到小依次为:乙醇浓度〉超声时间〉固液比〉超声功率,当乙醇浓度为30%、固液比为1∶10-1∶20、超声时间为30-60 m in、超声功率为600 W时,总黄酮提取率较高;优化后的提取工艺为:乙醇浓度40%,固液比1∶20,超声时间60 m in,超声功率600 W,此条件下总黄酮提取率为5.886%。[结论]牡荆总黄酮具有较强的抗氧化活性,且在0-0.125 5范围内具有较好的量效关系。 [ Objective ] The aim was to study the optimum extracting process of total flavonoids from Vitex quinata and its antioxidant activity. [ Method] With V. quinata leaf as the material, the single factor and orthogonal tests were taken to study the effects of ethanol concn., solidliquid ratio, ultrasonic time, ultrasonic power on extraction rate of total flavonoids. On the basis of it, the extraction process was optimized. With rat liver homogenate as the material, the antioxidant activity of total flavonoids from V. quinata was determined. [ Result ] The effects of all factors on extraction rate of total flavonoids from bid to small in order was ethanol concn. 〉 ultrasonic time 〉 solid-liquid ratio 〉 ultrasonic power. When ethanol was 30%, solid-liquid ratio was 1:10 - 1:2.0, ultrasonic time was 30 -60 min and ultrasonic power was 600 W, the extraction rate of total flavonoids was higher. The optimized extraction process was that: ethanol of 40%, solid-liquid ratio of 1:20, ultrasonic time of 60 rain, ultrasonic power of 600 W, and under the situation the extraction rate of total flavonoids was 5. 886%. [ Conclusion] The total flavonoids from V. quinata had stronger antioxidant activity and there was a good dose-effect relationship when its concn, was in 0 -0. 125 5 mg/ml.
