摘要
[目的]优化夏枯草多糖的脱色条件。[方法]以夏枯草为材料,经热水浸提,得到夏枯草提取液。研究离子交换树脂(D001、D315)和大孔吸附树脂(HZ-801、HZ-806、HZ-820)对夏枯草多糖的脱色效果。通过正交试验确定了夏枯草多糖脱色的最佳条件。[结果]HZ-801、HZ-806、HZ-820脱色效果好于D001、D315,脱色率、总糖保留率、蛋白质去除率分别在71%、54%、35%以上。影响夏枯草多糖脱色率的因素依次为:树脂种类>pH值>温度。pH值对总糖保留率和蛋白质去除率的影响最大,树脂种类对总糖保留率的影响最小,温度对蛋白质去除率的影响最小。[结论]夏枯草多糖最佳脱色条件为HZ-820树脂、pH值4.5、温度35℃。在此条件下,脱色率、总糖保留率和蛋白质去除率分别为86.77%、60.37%、89.73%。
[ Objective ] The aim was to optimize the deeoloring conditions for Prunella vulgaris polysaccharide. [ Method ] With P. vulgaris as the material, its extracting solution was obtained after extracted by hot water. The deeoloring effects of ion exchange resin( D001, D315 )and macroporous adsorption resin( HZ-801, HZ-806, HZ-820)on P. vulgaris polysaccharide were studied. And the optimum decoloring conditions for P. vulgaris polysaccharide was determined through orthogonal test. [ Result] The deeoloring effect of HZ-801, HZ-806, HZ-820 was better than that of D001 and D315, by which the deeoloring rate, total sugar retention rate, protein removal rate were above 71% , 54% and 35% resp. The influencing factors on decoloring rate of P. vulgaris polysaccharide in order was resin species 〉 pH value 〉 temperature. The effects of pH value on total sugar retention rate and protein removal rate were the biggest and the effect of resin species on total sugar retention rate was the smallest and the effect of temperature on protein removal rate was the smallest. [ Conclusion] The optimum decoloring condtions for P. vulgaris polysaccharide was that resin of HZ-820, pH value of 4.5, temperature of 35 ℃. Under this condition, the decoloring rate, total sugar retention rate, protein removal rate were 86.77%, 60.37% and 89.73% resp.
