云南昆明地区乙型肝炎病毒基因亚型与血清亚型研究

Distribution of subgenotypes and serotypes of hepatitis B virus in Yunnan province

目的研究云南乙型肝炎(HB)病毒基因亚型与血清型的分布情况。方法对云南97例乙型肝炎病毒(HBV)毒株S基因进行序列测定,通过Genebank HBV基因型分析网站确定毒株的基因型。通过序列比较、基因进化树的构建和相应位点特异氨基酸和核苷酸确定云南HBV毒株基因亚型和血清亚型。结果在所测的97例标本中,云南流行的HBV毒株有B基因型(75%)和C基因型(25%),没有发现其它基因型和基因混合型。B基因型有血清型adw2(占88%)和ayw1(12%);C基因型中以adrq+(96%)为主,仅1例为adw2(4%)。云南HBV B基因存在B2、B1、B4和B5亚型,其中以B2亚型为主,C基因型全为C2亚型。云南HBV C基因型毒株有其特异的核苷酸分布位点。结论云南HBV B基因型毒株与国内外参考株在S基因区没有显著的特异位点核苷酸分布,而C基因型则有显著的自身特征。

objective To study the distribution of the HBV subgenotypes and serotypes in Yunnan province. Methods Sequences of 97 surface antigen genes were used to assess the genotypes of the obtained strains. Subgenotypes and serotypes were determined based on phylogenetic analysis and the characteristic nucleotides and amino acids in the S gene. Results Both genotype B（75% ） and C（25% ） were found in this region. Apart from the described subgenotypes C2 and B2 in this region, B1, B4 and B5 also existed in our study supported by significant bootstrap values. Subgenotype B included serotype adw2 （ 88% ） and aywl （ 12% ）. In subgenotype C, serotype adrq ＋ accounted for 96% and serotype adw2, 4%. Conclusion Both genotype B2 and C2 are prevailed in Yunnan province although subgenotype B1, B4 and B5 are also detected in this study. All of serotype adw2, ayw1 and adrq ＋ are determined in this region.
