丹参及β-七叶皂甙钠减轻烫伤大鼠氧自由基损害的实验研究 被引量:2

Experiment Study of Mitigating the Injury Caused by Oxygen free Radicals with Salviae Miltrorrhizae andβ-aescinom Natricum in Burn Injury of Rats

摘要 目的:探讨丹参及β-七叶皂甙钠减轻大鼠烫伤后氧自由基损害的治疗作用及其机制,为临床治疗烧伤后急性肾损伤(Acute Renal Injury,ARI)提供理论依据。方法:制作30%TBSAⅢ度大鼠烫伤模型,并随机分为盐水组(M)、丹参组(S)、β-七叶皂甙钠组(A)及联合组(C,丹参+β-七叶皂甙钠),另设假烫对照组(N),每组8只。烫伤后24h取外周血测白细胞粘附聚集(LAA),取肾组织测丙二醛(MDA)、超氧化物歧化酶(SOD)及肾组织含水量(WCT)。结果:丹参组、β-七叶皂甙钠组及联合组外周血LAA、肾组织MDA及WCT明显低于盐水组,联合组最低;SOD含量明显高于盐水组,联合组最高。结论:丹参及β-七叶皂甙钠通过减轻中性粒细胞(PMN)在肾内的聚集、粘附,减少肾组织内氧自由基的含量,提高抗氧化剂SOD的水平及减少肾含水量等机制,对烧伤后ARI有一定防治作用,联合应用效果更佳。 Objective: To explore the treating effect and mechanism of mitigating injury caused by oxygen free radicals with salviae miltrorrhizae and β- aescinom natricum in the burn injury of rats so as to provide a theoretical basis for clinical treatment of acute renal injury of burns injury. Method: Forty rats were randomly divided into sham scalding control group (N, n = 8), sodium chloride group (M, n = 8), salviae miltrorrhizae group ( S, n = 8), β- acscinom natricum group. (A, n = 8), and combination group, i. e. salviae miltrorrhizae and β- aescinom natricum ( C, n = 8). The rats in M, S, A and C groups were subjected to 30% TBSA of Ⅲ degree scald on the back, and all the rats were sacrificed at 24 PBH. The blood and kidney samples were collected from the rats at 24 PBH for the determination of leukocyte adhesiveness/aggregation (LAA) in peripheral blood and malondialdehyde (MDA) and superoxide dismutase (SOD) contents, and the ratio of wet to dry weights (WCT) of kidney tissue. Result: Compared with those in M group, the LAA in blood and the kidney tissue contents of MDA and WCT in S and A groups, and especially in C group, were decreased significantly, but the SOD content in kidney tissue increased obviously in S and A groups, the highest observed in C group. Conclusion: Postburn intra - kidney agglutination and aggregation of PMN and kidney injury caused by oxygen free radicals (OFRs) could be inhibited by either Salviae Mihrorrhizae or β- aescinom natricum. In addition, these agents could also increase the tissue content of antioxidant and decrease renal water content. The results indicated that all the agent used might be effective in prevention and treatment of postbum renal injury, especially when used together.
