加味四物汤对荷骨肉瘤小鼠肿瘤VEGF及受体表达的影响 被引量:3

Effects of Modified Siwu Decoction on Expressions of VEGF and its Receptors in Osteosarcoma-bearing Mice

摘要 目的:探讨加味四物汤对对荷骨肉瘤小鼠肿瘤VEGF及其受体表达的影响。方法:建立KHOS成骨肉瘤原位移植模型,通过加味四物汤灌胃干预动物模型,采用RT-PCR方法定量检测移植瘤血管内皮细胞生长因子(VEGF)及其受体KDR和Flt-1的表达。结果:加味四物汤高剂量组有明显抑制肿瘤组织VEGF、Flt-1、KDR表达的作用(P<0.05);中、低剂量组抑制肿瘤组织VEGF、Flt-1、KDR表达的作用不明显(P>0.05);中药高剂量组抑制肿瘤组织VEGF、Flt-1、KDR表达的作用优于中、低剂量组(P<0.05)。结论:加味四物汤通过抑制VEGF、KDR、Flt-1的表达控制肿瘤新生血管的生长而达到阻止恶性骨肿瘤体内生长、转移的目的,但与药物浓度相关。 Objective:To investigate the effects of modified Siwu decoction (MSD) on the expressions of VEGF and its receptors in osteosareoma--bearing mice. Methods: After the orthotropie osteogenic sarcoma models were established, oral administration of MSD was applied to the mice. RT-PCR was performed to detect the expressions of VEGF, KDR and Flt --1 in tumor tissue. Results: High--dose MSD significantly suppressed the expressions of VEGF, KDR and Flt--1 in transplantable tumor tissue (P〈0.05), while the same effect was not observed in mid--dose and low-dose groups (P〈0. 05). The effect of inhibiting expressions of VEGF, KDR and Flt-1 in high-dose MSD was markedly superior to that in the mid--dose and low-dose MSD (P〈0.05). Conclusion: MSD can inhibit tumor angiogenesis via down-regulating the expressions of VEGF, KDR and Flt-1 thus blocking tumor survival and invasion. However, the effect is dose dependent.
