摘要
运用均匀设计法对超声波破碎根结线虫天敌真菌-1(destroying fungi of root-knotnematode以下简称DFRKN-1)细胞提取碱性磷酸酶的条件进行优化,摸索出了超声波破碎条件为:超声功率100 W,菌液浓度30%,超声时间2 s,间歇时间7 s,总时间35 min时,得到细胞破碎混合物中蛋白浓度最高,蛋白浓度从0.7306 g/mL提高到了2.6269 g/mL,比优化前提高了3.6倍;超声功率100 W,菌液浓度30%,超声时间6.5 s,间歇时间7 s,总时间为10 min时,得到细胞破碎混合物中碱性磷酸酶酶活力最高,从458 U/mL提高到1 725 U/mL,比优化前提高了3.8倍。
