小鼠淋巴瘤细胞Tk基因杂合性分析

摘要 目的研究小鼠淋巴瘤细胞L5178/Tk+/-3.7.2CTk位点杂合性,探讨Tk+/-基因稳定性,为实验检测过程中的质量控制提供可行的技术指标和实验依据。方法PCR分析L5178YTk+/-3.7.2C小鼠淋巴瘤细胞Tk位点的扩增产物。结果扩增产物为单一带型,未发现细胞株Tk杂合性丢失。结论被测试细胞株没有发生基因内和基因间突变、缺失、重组和移位等事件,说明细胞的遗传特性是稳定的。 [ Objective] To study the heterozygosity of Tk site on L 5178/Tk ^+/-3.7.2 C mouse lymphoma cells, investigate the genetic stability of Tk^+/- gene, and provide a feasible experimental evidence and guideline. [ Methods] Amplified products in Tk site of L 5178/Tk^+/-3.7.2 C mouse lymphoma cells were analyzed by PCR. [ Results] The amplified products were single band, and loss of Heterozygosity on Tk was not found. [ Conclusion ] The events such as intragenic and intergenic mutation, deletion, rearrangement and translocation do not occurred in the strain; it is suggested that the cells remained genetic stability.
