11β-羟类固醇脱氢酶1在代谢综合征大鼠模型中的表达及意义

摘要 目的探讨11β-羟类固醇脱氢酶1在代谢综合征大鼠模型中的表达及意义。方法健康雄性大鼠20只,随机分为2组,普通膳食对照组(A组)及高盐高热量膳食组(B组),喂养期间每周测一次进食量、体重和血压,符合代谢综合征模型标准后,选择终止日期,禁食12小时,用自动生化分析测量空腹血糖、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C),并将大鼠处死进行解剖,收集脂肪组织行RT-PCR检测11-βHSD 1mRNA表达的相对量,并进行统计学分析。结果高盐高热量膳食组体重、血脂、血糖、血压、11-βHSD1mRNA水平均较对照组增高,差别具有统计学意义(P<0.01)。结论高盐高热量饮食可引起11-βHSD1增加,11-βHSD1可能是引起代谢综合征特征的关键病因。 Objective To investigate the role and significance of 11-β hydroxysteroid dehydrogenase type 1 in a rat model of metabolic syndrome. Methods 20 Male 2-month-old Wistar rats were randomly divided into two groups: A for general dietary control group( n = 10);B for high-calorie, high-sah diet group( n = 10). Food intake, body weight and blood pressure were measured one time every week. When the models were finished, the date of termination was chosen. Fasting for 12 hours, tail vein blood were obtained, fasting blood glucose, triglycerides (TG), total cholesterol (TC), high density lipoprotein (HDL-C), low density lipoprotein cholesterol (LDL-C) were measured using automatic biochemical analyzer, rats were sacrificed autopsy, fat tissue was collected for testing expression of 11β-HSDlmRNA by RT-PCR.Results The levels of weight, blood lipid,blood glucose and 1lβ-HSD1 mRNA in Group B were statistically significant different to the control group A ( P 〈 0.01). C.ondusion The expression of 11β-HSD1 increased in group high-calorie, high-salt diet group, 11β-HSD1 might be the important factor in the metabolic syndrome.
