地黄饮子含药脑脊液对受损PC12细胞钙离子内流的影响 被引量:12

摘要 目的:探讨古方地黄饮子含药脑脊液对β-淀粉样蛋白(Aβ25-35)所致PC12细胞损伤的保护作用。方法:依据中药脑脊液药理学的方法,把PC12细胞分为空白组,模型组,西药对照组,地黄饮子脑脊液低、中、高剂量组六组,分别加入正常培养液、正常脑脊液以及含药脑脊液中,孵育后,加入钙离子荧光剂flou-3/AM负载后,于激光共聚焦显微镜下观察钙离子在细胞内的含量。结果:含有脑脊液各组加入Aβ25-35后虽然荧光强度升高,但荧光强度达到峰值的平均强度与模型组比较明显降低,差异有统计学意义(P<0.01),而且中、高剂量组与西药对照组比较,荧光强度降低,差异有统计学意义(P<0.05)。结论:地黄饮子脑脊液可以降低Aβ引起的钙离子内流。 objective:To discuss the cerebrospinal fluid with ancient prescriptions Dihuangyinzi which has protective function on PC12 cells,injured by β-amyloid protein(A β25-35).Methods: We divide PC12 cells into 6 groups—that is,the blank group,the model group,the control group of western medicine,and the cerebrospinal fluid groups with low-dose,medium-dose and high-dose of Dihuangyinzi with pharmacology method of cerebrospinal fluid containing Chinese medicine;and add normal culture medium,the normal cerebrospinal fluid and cerebrospinal fluid containing drugs respectively;after incubation,then add calcium fluorescent agent flou-3/AM and observe the content of calcium in the cell under the laser scanning microscope after load.Results: Although fluorescence intensity in each cerebrospinal fluid group increases after adding A β25-35,the average strength of the fluorescence intensity reaching its peak decreases significantly when compares with the model group;the difference has statistics significance(P<0.01),and the fluorescence intensity of medium and high groups decreases when compares with western medicine control group,and the difference has statistics significance(P <0.05).Conclusion: Cerebrospinal fluid with Dihuangyinzi can reduce Calcium ion,induced by Aβ,internal flowing.
