玉屏风散提取液对小鼠脾淋巴细胞增殖及转化的影响

Effect of jade screen powder extracting solution on proliferation and ransformation in splenic lymphocytes of mice

目的研究玉屏风散提取液对小鼠脾淋巴细胞增殖、转化的影响。方法分别制备玉屏风散水煎液、玉屏风散蒸馏水超声提取液、玉屏风散无水乙醇超声提取液和玉屏风散乙酸乙酯超声提取液。分离健康昆明种小鼠原代脾淋巴细胞进行体外培养,培养基中分别加入不同剂量上述药液使终末浓度达到1.67 mg.mL-1、3.33 mg.mL-1、8.33 mg.mL-1、16.67 mg.mL-1(折合生药)进行细胞培养,以0 mg.mL-1组为对照组。培养72 h后加入2-(2-甲氧基-4-硝基苯基)-3-(4-硝基苯基)-5-(2,4-二磺酸苯)-2H-四唑单钠盐(WST-8)用酶标法分别检测4种玉屏风散提取液对脾淋巴细胞增殖和刀豆蛋白A(ConA)诱导的脾淋巴细胞转化的影响。结果与对照组相比,玉屏风散(1.67 mg.mL-1、3.33 mg.mL-1、8.33 mg.mL-1、16.67 mg.mL-1)可显著促进小鼠脾淋巴细胞增殖和ConA诱导的脾淋巴细胞转化(P<0.01)。除1.67 mg.mL-1玉屏风散乙酸乙酯超声提取液外,其他剂量的玉屏风散超声提取液促进小鼠脾淋巴细胞增殖和ConA诱导的脾淋巴细胞转化的效果均低于同剂量组的玉屏风散水煎液(P<0.05)。结论玉屏风散可显著促进小鼠脾淋巴细胞增殖和ConA诱导的脾淋巴细胞转化,但玉屏风散的超声提取条件需进一步摸索。

Objective To investigate the effect of jade screen powder extracting solution on proliferation and transformation in splenic lymphocytes of mice.Methods Prepared water solution,supersound extract of distilled water,supersound extract of dehydrated alcohol and supersound extract of acetidin of jade screen powder,and separated primary splenic lymphocytes from healthy Kunming mice to culture in virto.Different concentrations（1.67 mg·mL^-1,3.33 mg·mL^-1,8.33 mg·mL^-1,16.67 mg·mL^-1） of jade screen powder were added to（convert into crude drug） in nutrient medium to culture the cells respectively.0 mg·mL^-1 group was set up as control group.4-[3-（4-nitrophenyl）-2-（2-methoxy-4-nitrophenyl）^-2H-5-tetrazolio]-1,3-benzene disulfonate（WST-8）was added to nutrient medium after culturing for 72 h.The proliferation and transformation induced by ConA of splenic lymphocytes were detected by enzyme linked immunosorbent assay.Results Compared with the control group,jade screen powder groups（1.67 mg·mL^-1,3.33 mg·mL^-1,8.33 mg·mL^-1,16.67 mg·mL^-1） promoted the proliferation and transformation of splenic lymphocytes which induced by ConA significantly（P〈0.01）.Except supersound extract of acetidin of jade screen powder（1.67 mg·mL^-1）,the effects of other concentrations of supersound extract were lower than the same concentration of decoctum of distilled waters（P〈0.05）.Conclusion Jade screen powder can promote the proliferation and transformation of splenic lymphocytes which induced by ConA significantly,but selective condition of supersound extraction of jade screen powder should be further explored.
