高效液相色谱法测定不同产地白芷中欧前胡素和异欧前胡素含量

Determination of contents of imperatorin and isoimperatorin in Radix Angelicae Dahuricae from different areas by HPLC

目的采用高效液相色谱法（HPLC）同时测定不同产地白芷中欧前胡素和异欧前胡素的含量,为更好的评价不同产地白芷的质量提供依据。方法色谱柱：L ichrospher-C18（250×4.6 mm,5μm）,柱温：30℃,流动相：甲醇-水,梯度洗脱,流速为1.0 mL·min^-1。结果欧前胡素、异欧前胡素进样量分别在0.54-5.40μg、0.244-2.44μg范围内,呈现良好的线性关系。欧前胡素平均回收率99.020%,相对标准偏差（RSD）3.318%;异欧前胡素平均回收率97.528%,RSD 2.904%。欧前胡素平均含量为1.465 mg·g^-1;异欧前胡素平均含量为0.575 mg·g^-1。结论该测定方法简单,快速,重现性好,可作为该药材质量的控制方法。

Objective To develop a method for the determination of imperatorin and isoimperatorin in Radix Angelicae Dahuricae.Methods A HPLC method was established.Lichrospher-C18 column（250 mm×4.6 mm,5 μm）was used,with mixture of methanol and water in a gradient mode.The flow rate was 1.0 mL·min^-1.Results The calibration of imperatorin was linear in range of 0.54～5.40 μg.The average recovery was 99.020%,RSD 3.318%.The calibration of isoimperatorin was linear in range of 0.244～2.44 μg.The average recovery was 97.528%,RSD 2.904%.The average contents of imperatorin and isoimperatorin were 1.465 mg·g^-1 and 0.575 mg·g^-1 respectively.Conclusion The method is appropriate for the detemination of Radix Angelicae Dahuricae.
