摘要
目的:观察普伐他汀对血管紧张素Ⅱ(AngⅡ)诱导大鼠血管平滑肌细胞(VSMCs)增殖的影响,以探讨他汀类药可能的非调脂抗动脉粥样硬化作用。方法:组织贴块法培养的大鼠VSMCs随机分为对照(正常VSMCs)组、AngⅡ(10-6mol/L)模型组、普伐他汀高剂量(10-5mol/L+AngⅡ10-6mol/L)、中剂量(10-6mol/L+AngⅡ10-6mol/L)、低剂量(10-7mol/L+AngⅡ10-6mol/L)组、溶剂(体积分数0.1%的DMSO+AngⅡ10-6mol/L)组,分别测定活细胞数量和细胞周期中各期细胞构成比。观察普伐他汀对VSMCs增殖和迁移的影响。结果:AngⅡ(10-6mol/L)作用24 h能使活细胞数量明显增加,并使VSMCs的G0/G1期细胞减少,细胞增殖指数增大。与AngⅡ模型组比,普伐他汀组活细胞数减少,G0/G1期的细胞构成比增加,细胞增殖指数减少。结论:AngⅡ对VSMCs有促增殖作用,能被普伐他汀所拮抗。
Objective: To investigate the effects of pravastatin on cell proliferation induced by angiotensin Ⅱ (AngⅡ) in vascular smooth muscle cells( VSMCs) of rats. Methods: The VSMCs cultured by tissue-sticking method were randomly divided into control, Ang IT model, pravastatin and solvent groups. MTT values and cell cycle distribution were determined. Results: Twenty-four hours exposure to AngⅡ (10^-6mol/L)caused an increase valube in MTT and the cell mitogenic activity was also increased. The cell proliferation was inhibited by pravastatin. Pretreatment with pravastatin increased percentages of G0/G1 period cells. The cell mitogenic activity induced by AngⅡ was aslo reduced. Conclusion: Pravastatin antagonizes the cell proliferation induced by AngⅡ in VSMCs of rats.
