兔辅酶Ⅱ依赖性视黄醇脱氢/还原酶多克隆抗体的制备

摘要 目的:制备兔辅酶Ⅱ依赖性视黄醇脱氢/还原酶(NRDR)多克隆抗体。方法:应用原核表达的兔NRDR免疫豚鼠,制备豚鼠抗兔NRDR多克隆抗体,并以Western blot法分析抗体特异性。结果:免疫豚鼠获得了高效价抗血清,效价为1∶2 000时检测极限为160 ng蛋白,与NRDR蛋白能特异性结合。结论:获得的多克隆抗体应用于Western blot中检测兔NRDR效果良好,为NRDR的进一步研究打下基础。 Objective: To prepare the polyelonal antibodies of rabbit NADP(H)-dependent retinol dehydrogenase/reductase(NRDR). Methods: The purified protein was used to immunize guinea pig to prepare polyclonal antibodies which were finally analyzed by Westem blot. Results: The immunized guinea pig produced high titer antiserums with a high specificity against NRDR. The detection limit was 160 ng NRDR protein with the titer of 1:2 000. Conclusion: The anti- NRDR polyelonal antibodies perform well in Western blot assay for NRDR and are supposed to facilitate the research of NRDR.
