摘要
马立克氏病病毒(MDV)被膜蛋白VP22被证实能在细胞间高效转导,为了进一步证明VP22能够作为蛋白转运的载体,将4种不同的异源蛋白与MDV1型(MDV-1)CVI988/Rispens株VP22蛋白融合表达,通过观察这些融合蛋白的细胞定位以及它们的细胞间扩散能力来研究VP22转运蛋白的情况。结果发现,禽流感病毒(AIV)核蛋白(NP)、牛γ干扰素(BoIFN-γ)及新城疫病毒(NDV)F蛋白能够被MDV-1VP22转运,而鸡法氏囊病病毒(IBDV)蛋白VP2不能被MDV-1VP22转运,上述结果说明MDV-1VP22蛋白能够作为蛋白转运的载体,但对所转运的蛋白具有选择性。
Marke's disease virus tugement protein VP22 is previously showed to possess a remarkable property of intercellar transport. To confirm whether it can be utilized as a cargo for protein delivery, we investigated subcellular localization of four heterogeneous proteins fused with the VP22. Diffusion of fusion proteins in monolayers was detected with specific monoclonal antibodies. The results showed that the nucleoprotein (AIV-NP) of avian influenza disease virus, bovine IFN- ~/ (BIFN-~,) and Newcastle disease virus F protein could be delivered by VP22 of strains CVI988/Rispens. However the IBDV VP2 protein could not be delivered. It suggested that MDV-I VP22 may be selective in its load while acting as a cargo in protein delivery.
