摘要 背景:瞬时感受电位(transient receptor potential,TRP)通道在人体遍布多种组织和细胞,并参与人体几乎所有的生理功能和许多病理变化。目的:观察瞬时感受电位基因亚家族成员TRPV4在大鼠睾丸组织中的表达。设计、时间及地点:单一样本观察。实验于2007-03/2008-04在广东省人民医院医学研究中心完成。材料:SD大鼠5只;TRPV4引物由上海生物工程技术服务有限公司合成。方法:选用SD大鼠,取20mg睾丸组织,用RNAprep_动物组织/细菌总RNA提取试剂盒提取总RNA,取1μg总RNA,用于合成cDNA。主要观察指标:用常规反转录-聚合酶链反应和免疫组织化学染色检测TRPV4在大鼠睾丸组织中的表达。结果:常规反转录-聚合酶链反应方法检测到在TRPV4的泳道中可见一条带,大小与期望长度符合;免疫组织化学染色可见TRPV4阳性片中较成熟的生精细胞和支持细胞为黄褐色。结论:TRPV4在睾丸组织上有表达,主要表达在较成熟的生精细胞和支持细胞上。 BACKGROUND: The transient receptor potential (TRP) protein channel distribute many kinds of tissue and cell in human body, which take part in all kind of physiologic function and pathological changes. OBJECTIVE: To study the expression of TRPV4, a subfamily member of TRP gene in rat's testis. DESIGN, TIME AND SETTING: A single sample observation was performed at the Research Center of Guangdong General Hospital from March 2007 to April 2008. MATERIALS: Five healthy male SD rats were collected in the study. The primer of TRPV4 was constructed by Shanghai Sangon Biological Engineering Technology & Services Co., Ltd. METHODS: A total of 20 mg testes tissues of rats were extracted total RNA with total RNA extraction kit, 1 μg total RNA was obtained and to synthesis cDNA. MAIN OUTCOME MEASURES: The expression of TRPV4 in testis was detected by RT-PCR and immunohistochemistry staining. RESULTS: A strap which in line with the expected length was detected in the track of TRPV4 by RT-PCR. Spermatogenic cell and Sertoli cell was stained cinnamomeous by Immunohistochemistry. CONCLUSION: TRPV4 expresses on the rats' testes, especially in spermatogenic cell and Sertoli cell.
