摘要
背景:骨关节炎的病理改变涉及多种关节构成组织,其中骨骼肌的地位日渐引起研究者关注。目的:通过观察骨关节炎C57小鼠骨骼肌收缩蛋白α-肌动蛋白与肌球蛋白重链基因表达状况,探讨柔肝中药软骨制剂利胶囊对收缩蛋白基因表达的影响。设计、时间及地点:随机对照动物实验,于2004-09/2007-01在上海市中医药研究院骨伤科研究所完成。材料:选取7周龄清洁级C57小鼠18只,雌雄各半,体质量(20±5)g。软骨利胶囊(柔肝中药制剂),由上海中医药大学中药标准化中心提供。方法:18只C57小鼠按随机数字表随机分为正常组、模型组、软骨利组,每组6只。除正常组外,对小鼠进行运动负荷训练造模。软骨利组给予0.2mL软骨利溶液(约含2mg软骨利)灌胃5周,正常对照组和模型组给予0.2mL生理盐水灌胃5周。主要观察指标:应用反转录-聚合酶链反应检测股四头肌肌α-actin、肌球蛋白重链4种亚型(Ⅰ,Ⅱa,Ⅱd/x,Ⅱb)mRNA表达水平。结果:与空白组比较,模型组α-actin mRNA显著低表达(P<0.05),肌球蛋白重链Ⅱa,肌球蛋白重链Ⅱd/x mRNA低表达(P<0.05)。与模型组比较,软骨利干预组α-actin与多种肌球蛋白重链亚型mRNA表达水平上调(P<0.05)。结论:柔肝中药制剂软骨利能够促进骨骼肌收缩蛋白基因表达,提示其可改善骨关节炎动物骨骼肌肌力。
BACKGROUND: Multi-tissues are involved in osteoarthritis pathology, and the effect o4 skeletal muscle in the pathogenesis has attracted more attention.
OBJECTIVE: To investigate the mRNA expression of α -actin, myosin heavy chain (MHC) isoforms in C57 mice with osteoarthritis, in addition, to explore the effect of Ruangufi (liver-softening formulation) on the treatment of osteoarthritis.
DESIGN, TIME AND SETTING: A randomized control animal experiment was performed at the Department of Orthopedics and Traumatology, Tradition Chinese Medicine Academy of Shanghai between September 2004 and January 2004.
MATERIALS: A total of 18 healthy, 7 weeks old, clean grade, C57 mice, with body weight of (20±5) g, half male and half female, were selected. Ruangufi was provided by Standardization Center of Tradition Chinese Medicine Academy of Shanghai.
METHODS: Eighteen C57 mice were randomly divided into blank control, model, and Ruangufigroups, with 6 animals in each group. All mice were prepared exercise load models exception of the blank control group. Mice in the Ruanguli group were intragastrically administrated 0.2 mL Ruanguli solution (containing 2 mg Ruanguh) for 5 weeks. Animals in the other two groups were administrated 0.2 mL physiological saline fro 5 weeks.
MAIN OUTCOME MEASURES: The mRNA expression level of a -actin and MHC isoforms in quadriceps femoris was tested by RT-PCR.
RESULTS: The level of α -actin mRNA in model group was lower than that of the normal control group (P 〈 0.05), with low expression of MHC Ⅱ a, and MHC Ⅱ d/x mRNA (P 〈 0.05). Compared with the model group, a -actin and MHC isoforms was up-regulated in the Ruanguli group (P 〈 0.05).
CONCLUSION: Ruangufi can promote the gene expression of contractile protein, which is capable for strength the myodynamia of skeletal muscle.
