摘要
采用石墨炉原子吸收光谱法,对6例接受大剂量卡铂化疗的肺癌患者首、末次给药时的血浆总铂浓度进行了监测,并对其药代动力学特点作了分析。结果:首、末次给药后15分血浆总铂峰值浓度分别为(14.73±7.26)mg/L和(18.01±7.79)mg/L,末次给药与首次给药相比,药时曲线下面积(AUC)增加(48.82±39.53)mg/(L.h),血清铂清除速率(CLs)下降(7.82±5.60)mg/h。并对参数改变的机理进行了初步探讨。
In this study, graphite furnace atomic absorption spectrophotometry was adapted to the plasma concentrations of total platinum of 6 patients with lung cancer at the first and last time infusions of high dose carboplatin chemotherapy,and the pharmacokinetic studies were also performed. The results showed that the first and last peak concentrations of total carboplatin in plasma were (14. 73 ± 7. 26) mg/L and (18. 01 ± 7. 79)mg/L at 15 min after infusions ended. Compared with the first time infu-sion,the area under the concentration curve(AUC)of the last time infusion increased (48. 82±39. 53) mg/(L · h)and the systemic clearance(CLs)decreased (7. 82±5. 60)mg/h. The mechanisms of these changes in pharmacokinetics were discussed in detail.
