反相高效液相色谱法同时测定熊胆丸中大黄素和大黄酚含量

摘要 目的建立高效液相色谱(RP-HPLC)法测定熊胆丸中大黄素和大黄酚的含量。方法色谱柱为HypersilC18柱(200mm×4.6mm,5μm),以甲醇-0.1%醋酸(75∶25)为流动相,流速为1.0mL/min,检测波长225nm。结果大黄素和大黄酚质量浓度线性范围分别为0.605~6.050μg/mL和2.04~20.40μg/mL,r=0.9999(n=5),平均加样回收率分别为99.40%和100%,RSD分别为1.83%和0.91%。结论该方法准确、简便、专属性强,可用于控制熊胆丸的质量。 Objective To establish a HPLC method for the determination of emodin and chrysophanol in Xiongdan Pill. Methods HPLC was adopted. Emodin and chrysophanol were simuhaneously separated and determined on the Hypersil C18 column (200 mm ×4.6 mm, 5 μm). The mobile phase consisted of methanol-0. 1% acetic acid (75:25). The flow rate was 1.0 mL/min, the determination wavelength was 225 nm. Results Their linear ranges were 0. 605-6. 050 μg/mL for emodin, 2.04-20. 40 μg/mL for chrysophanol. Their correlation eoeff- eients were both 0. 999 9. Their average recovery rates were 99.40% and 100%, RSD were 1.83% and 0.91%, respectively. Conclusion The method is proved to be simple and accurate, and can be used for the quality control of this product.
