摘要
应用荧光光谱和紫外-可见光谱研究了卟啉锌(HP—Zn)与牛血清白蛋白(BSA)的相互作用及在超声波作用下HP—Zn对BSA的损伤作用,并探讨了超声照射时间,HP—Zn浓度和溶液酸度等因素对BSA损伤的影响。结果表明,37℃时,BSA与HP—Zn的结合数n=1.0379,其平衡常数KA=6.2661×10^4,两者之间的作用距离r=3.53nm。在一定条件下,BSA的损伤程度随着超声照射时间.HP—Zn浓度和溶液酸度的增大而增大。这一结果对声动力学疗法(SDT)治疗肿瘤应用于临床具有重要的意义。
The interaction between hematopotphyrin zinc (HP-Zn) and bovine serum albumin (BSA) and damage of BSA in the presence of HP-Zn under ultrasonic irradiation were studied by UV -Vis and fluorescence spectra. The influences of ultrasonic irradiation time, HP-Zn concentration and solution acidity on damage of BSA were also detected. The results showed that at 37℃, the binding site number (n), the apparent binding constant (KA) and the binding distance (r) were 1. 0379, 6. 2661 × 10^4and 3.53 nm, respectively. Under different conditions, the damage degree of BSA rose up with the increasing of ultrasonic irradiation time, HP-Zn concentration and solution pH value. The research results are of great significance for driving sonodynamic treatment (SDT) to clinic application.
