摘要
目的:研究小鼠肝脏内Mac-1阴性自然杀伤细胞分泌细胞因子的能力。方法:采用poly I:C和IL-2分别在体内和体外激活小鼠肝脏内的NK细胞,采用尼龙毛柱和免疫磁珠分选提纯Mac-1阴性和阳性自然杀伤细胞,用酶联免疫斑点(ELISPOT)法检测IFN-γ、IL-4、IL-5三种细胞因子的分泌情况。结果:Mac-1阴性自然杀伤细胞优势分泌IL-4、IL-5;Mac-1阳性自然杀伤细胞优势分泌IFN-γ。结论:Mac-1阴性自然杀伤细胞属于NK2细胞亚群;Mac-1阳性自然杀伤细胞属于NK1细胞亚群。
Objective:To study the cytokine secretory capacity of Mac-1 negative natural killer cell in mouse liver.Methods:NK cells in mouse liver were activated by poly I:C and IL-2 both in vivo and in vitro.Mac-1 negative and positive natural killier cells were purified with nylon wool and MACS system cell sorting,and secretion of IFN-γ,IL-4 and IL-5 was determined using enzyme-linked immunosorbent Dot(ELISPOT).Results:Mac-1 negative natural killer cells secreted IL-4 and IL-5;and Mac-1 positive natural killer cells produced IFN-γ upon activation.Conclusion:Mac-1 negative natural killer cells are in NK2 cell subsets:whereas Mac-1 positive natural killer cells are in NK1 cell subsets.
