HPLC法同时测定玉屏风散中色原酮、黄酮和内酯类9种成分的含量 被引量:9

Simultaneous Determination of Four Chromones,Three Flavonoids and Two Atractylenolqides in Yupingfeng Powder by HPLC

摘要 目的建立以高效液相色谱法同时测定玉屏风散中3类化合物(黄酮类、色原酮类和内酯类)9种有效成分:升麻素苷、毛蕊异黄酮苷、升麻素、5-O-甲基维斯阿米醇苷、亥矛酚苷、毛蕊异黄酮、芒柄花素、白术内酯III和白术内酯I含量的分析方法。方法采用KromasilC18ODS柱(4.6mm×250mm,5μm);柱温30℃;流动相为乙腈-0.15%(V/V)磷酸水溶液,梯度洗脱;流速1mL/min;检测波长220nm。结果9种成分在各自的线性范围内,标准曲线呈良好的线性关系,相关系数均在0.9998以上;平均加样回收率分别为104.0%,100.7%,99.6%,99.1%,98.6%,96.0%,100.0%,99.4%,101.8%,RSD值均小于3%。结论该方法简单、准确、重现性好,可为玉屏风散的质量监控及新剂型研究提供更加全面、可靠的检测方法。 Objective To develop a HPLC -based method for simultaneous determination of four chromones, three flavonoids and two Atractylenolqides including prin - 0 - glucosylcimifugin, calycosin - 7 - 0 - β - D - glucopyra- noside, cimifugin, 4' - O - β - D - glucosyl - 5 - 0 - methylvisamminol, sec - 0 - glucosylhamaudol, calycosin, formononetin, atractylenolide III and atractylenolide I in Fupingfeng Powder. Methods These nine compounds were an- alyzed simultaneously with a Kromasil Cl80DS column (4.60 mm × 250 mm, 5 μm) by gradient elution, and 0. 15 % ( V/V) phosphoric acid - acetonitrile used as the mobile phase. The column temperature was maintained at 30℃, the flow rate was 1 mL/min, and the detection wavelength was set at 220 nm. Results All nine compounds showed good linearity in the range of the test concentration, with correlation coefficients over 0. 999 8. The average recoveries of the nine com- poundswerel04.0%, 100.7%, 99.6%, 99.1%, 98.6%, 96.0%, 100.0%, 99.4% andlO1.8%, respec- tively. Conclusion The method has the advantages of simplicity, precision and reliability, and can provide a more comprehensive and reliable way for the quality control of Yupingfeng Powder and the development of its new related preparations.
