汉防己对胸腺细胞Fas和Fas-L表达的影响 被引量:1

摘要 目的探讨防己(Radix Stephaniae Tetrandrae)对小鼠胸腺的细胞的影响及其作用机制。材料与方法取昆明小鼠的胸腺细胞,随机分为阴性实验组(无防己)、阴性对照组(用0.05MPBS代替一抗)和3个药物实验组(其浓度分别为10μg/ml、50μg/ml、100μg/ml),分别培养1h、6h、12h、24h后,利用免疫细胞化学的方法检测细胞内Fas和Fas-L的阳性细胞的表达。结果随着汉防己煎剂浓度的增高及作用时间的延长,实验组Fas和Fas-L的阳性细胞表达率显著增高(P<0.05)。结论防己可以诱导小鼠胸腺的细胞的凋亡。 Objective To investigate the effect of radix stephaniae tetrandrae on the thymocyte.Methods The cultured thymic cells of Kunming mice were randomly divided into negative experi- ment group, negative control group(PBS instead of Ist antibaody) and 3 drug group, which were treated by 10 μg/ml, 50 μg/ml and 100 μg/ml Radix Stephaniae Tetrandrae decoction, respective- ly. The expression of Fas and Fas-L were observed by immunocytochemical method after 1 h, 6 h, 12 h and 24 h treatment. Results The expressional rates of Fas and Fas-L increased significantly with the action time and enhancing the drug concentration, compeared to the control group (P〈0.05). Conclusion The Radix Stephaniae Tetrandrae decoction could induce the apoptosis of mice thymocytes.
