温州市瓯海地区汉坦病毒的分子流行病学调查

The molecular epidemiological investigation of hantavirus in Ouhai District of Wenzhou City

摘要 目的研究温州市瓯海地区宿主携带汉坦病毒的状况和型别。方法经间接免疫荧光(IFA)试验初筛为阳性的鼠肺标本,用巢式RT—PCR扩增阳性标本中汉坦病毒的S基因片段上的目的核苷酸序列(620~999nt),并以ClustalX(1.831和Phylip3.63构建系统进化树进行分型和分子进化分析。结果温州市瓯海地区共捕获啮齿动物148只,5份鼠肺标本中汉坦病毒抗原阳性,其中褐家鼠3只,黄胸鼠2只,病毒携带率为3.38%,与汉坦病毒分型标准比较,均属于SEOV型。结论瓯海地区褐家鼠和黄胸鼠携带S1和S3亚型汉坦病毒,显示出汉坦病毒的多样性。 Objective TO gain more insight into the staus of hantavirus (HV) infection and their genotype in rodents in Ouhai District of Wenzhou City of Zhejiang. Methods The partial S fragment (620-999nt) from the HV antigen positive rodents lungs detected by indirect immunofluoresence assay was amplified with nested RT-PCR using genetype-specifie primers,the phylogenetic analysis was performed with ClustalX (1.83) and Phylip3.63 software. Results A total of 148 rodents were trapped in Ouhai District, HV antigen were identified in 5 out of these rodents lungs,3 of which from Rattus norvegicus and 2 from Ratttts flavipectus,and the overall prevalence of HV in rodents was 3.38%. Conclusion The main genotypes of HV carried by Rattus norvegicus and Rattus flavipectus in Ouhai District are genotypes S1 and S3 SEOV, and the HV type is diverse in Ouhai District.
