乳酸杆菌SD-22产类细菌素发酵条件的优化 被引量:11

Optimization of Fermentation Conditions for Production of Bacteriocin-like Substance by Lactobacillus Brevis SD-22

摘要 为了获得最大量的类细菌素,通过单因素和正交试验优化乳酸杆菌SD-22产类细菌素的营养条件和环境条件。结果表明:产类细菌素的最佳培养条件为30℃厌氧培养48h,培养基初始pH6.5,接种量1%,种龄12h。培养基组分的最佳组合为大豆蛋白胨15g、酵母粉15g、牛肉膏5g、葡萄糖2g、K2HPO42g、柠檬酸氢二铵2g、乙酸钠5g、MgSO4·7H2O0.58g、MnSO4·4H2O0.25g、吐温-802mL,用蒸馏水定容至1000mL。在此条件下培养乳杆菌SD-22,抑菌圈直径可达20.12mm。 In order to maximize the yield of bacteriocin-like substance by liquid cultivation of Lactobacillus brevis SD-22, single-factor-experiment design and Ortho-experiment design were used to optimize the environmental and nutrient conditions. The optimal conditions were obtained at incubation time of 48 h, 30 ℃,initial pH 6.5,inoculation rate of 1% and seed age of 12 h. The optimum nutrients in the media were soybean peptone 15 g, bear extract 5 g, yeast powder 15 g, glucose 20 g, K2HPO4 2 g, NaAc 5 g, MgSO4·7H2O 0.58 g, Di-Ammonium citrate 2 g, MnSO4·4H2O 0.025 g, Tween-80 2 mL, and water 1000 mL. The diameter of inhibitory activity of Lactobacillus brevis under the optimum broth was 20.12 mm.
