摘要
目的和方法:应用免疫细胞化学方法,观察穿心莲成分API0134对轻度氧化低密度脂蛋白(mmLDL)诱导人血单核细胞表达血小板源生长因子B链蛋白(PDGF-B)和碱性成纤维细胞生长因子(bFGF)的影响;并进行平滑肌细胞(SMC)有丝分裂试验,观察API0134作用后的单核细胞条件培养基(CM)对3H-TdR掺入SMCDNA的影响。结果:mmLDL可显著促进单核细胞表达PDGF-B和bFGF,mmLDL作用后的CM亦可显著增加3H-TdR的掺入量。mmLDL的上述作用可被不同剂量的API0134和API0134作用后的CM所抑制。结论:API0134的作用可能是抗动脉粥样硬化的机理之一。
AIM and METHODS:The effects of API0134 on expression of platelet-derived growth factor B chain protein(PDGF-B) and basic fibroblast growth factor(bFGF) in human monocytes induced by slight-oxidative modified low density lipoprotein(mmLDL) were studied by immunocytochemical methods,and the effects of monocyte conditioned media(CM) on 3H-TdR in corporation into DNA of smooth muscle cell(SMC) were observed. RESULTS: mmLDL induce the expression of PDGF-B and bFGF in monocytes significantly, and 3H-TdR incorporation into DNA of the SMC were also promoted significantly by the CM treated with mmLDL. The inductive effect of mmLDL can be inhibited by API0134 as well as by CM treated with API0134. CONCLUSION:The inhibitory effects of API0134 may be one of its anti-atherosclerotic mechanisms..
