眼镜蛇毒直接溶解因子对人肝癌细胞株H_(7402)产生NO和LPO的影响 被引量:12

摘要 目的:研究眼镜蛇毒直接溶解因子(DLF)对人肝癌细胞株H7402产生一氧化氮(NO)和脂质过氧化物(LPO)的影响。结果:DLF对体外培养的H7402细胞有杀伤作用,IC50为247mg·L-1,DLF与H7402细胞的生长抑制率呈量效关系。DLF可介导H7402细胞产生大量NO2,NO2量与DLF的剂量呈量效关系。在一定剂量范围内NO2量与H7402的生长抑制率呈显著正相关。用570型粘附式细胞仪测定单个细胞内LPO,发现DLF可使H7402细胞内LPO产生增加。结论:DLF可杀伤H7402细胞,其部分机制可能是通过诱导肿瘤细胞产生NO和LPO。 AIM:To examine the effect of direct lytic factor (DLF) from Naja naja atra venom on nitric oxide (NO) and lipid peroxides (LPO) produced by human liver cancer cell line H 7402 RESULTS: DLF was cytotoxic on H 7402 cells cultured in vitro ,IC 50 was 2.47mg·L -1 .Growth-inhibiting rate of H 7402 in response to DLF was dose-dependent. As a stable metabolite of NO, nitrite production in culture supernatants was used to represent the level of NO. A large amount of NO 2 was produced by H 7402 cells which were cultured with DLF, production of NO 2 by H 7402 cells in response to DLF was dose-dependent. Close positive correlation was found between the level of ON 2 and growth-inhibiting rate of H 7402 in a certain range. Measuring the level of LPO in single cells production of LPO in H 7402 cells was increased by DLF CONCLUSION:Human liver cancer cell line H 7402 was damaged by DLF, the mechanism might be partly due to inducing the tumor cells to produce NO and LPO. .
