Abstract It has been reported that the adrenoceptors were desensitized following shock. The adrenoceptor's desensitization included the receptor number downregulation, decreased receptor affinity, and altered enzyme system which is coupled with these receptors. There are several reasons for the adrenoceptors desensitization during circulatory shock. First, high concentration of catecholamines released following shock might induce adrenoceptor phosphorylation and internalization, which could make the surface receptors become sequestered. Second, endotoxin and mediators such as tumor necrosis factor could cause the decrease of adrenoceptor number, affinity and responsiveness. Third, a long period of severe ischemia and hypoxia also impairs the functions of adrenoceptor system during shock, in addition, endogenous opioid peptides released during shock inhibit the adrenoceptor system.
