摘要
本文采用放射酶分析法检测红细胞胰岛素降解酶活性(EIDEA)。该方法应用方便、灵敏度极高、结果准确。并对酶反应最佳条件进行了实验探讨,底物Km为0.09mU/ml,最适pH值7.40,延滞期25分钟,25~40分钟测得BIDEA基本一致。批内CV为3.0的,批间CV为3.3%。氯化物、EDTA可抑制该酶活性。50例健康人红细胞胰岛素酶活性为80,18±13.80U/mgHb,无性别、年龄差异。
This paper described the method of radioassay of insulin. degrading enzyme activity of erytrocytes(EIDEA) It is simple, sensitive and accurate Optimum reaction conditions for EIDEA were investigated, The substrate Km was 0.09mU/ml. The optimum pH was 7.40.The delay time wits 25 minutes. The results of dctcrmination from 25 to 40 minutes were basically identical Within - run and between - run preeision were 3.0% and 3.3%,respeetively. Chloride and BDTA can inhibit EIDBA. The mean EIDEA of 50 healthy subjects was 80.18±13.80mU/mlHb.There was no sex or age difference.
