长链酰基辅酶A合成酶(LACSs)把游离脂肪酸活化成为酰基辅酶A硫脂,在油脂的合成和降解途径中发挥着重要的作用。通过电子克隆的方法,发现了1个油菜(Brassica napus)LACS基因。该基因长2270 bp,包含2004 bp的开放阅读框,预测编码1个含有667个氨基酸残基的蛋白质,命名该蛋白为BnLACS4,序列分析显示,BnLACS4具有典型的LACS分子特征。表达分析显示BnLACS4在成熟油菜的根、茎、叶和花中都有表达。在授粉后(DAP)35 d,不同含油量的角果皮和种子中,BnLACS4的表达也存在差异。表达谱说明,BnLACS4可能参与油脂的生物合成以及油菜种子中的油脂积累。
Long chain acyl-coenzyme A synthetases (LACSs) activate free fatty acid to acyl-CoA thioesters, and play important roles in the biosynthesis and degradation of lipids. In this study, a Brassica napus LACS gene was found through in silicon cloning. The gene is 2 270 bp with 2 004 bp of open reading frame (ORF) , encoding a protein consisting of 667 amino acid residues which was designated BnLACS4. Sequence analysis indicated that BnLACS4 possessed typical molecular characteristics of LACS. Expression analysis revealed, BnLACS4 was expressed in mature roots, stems, leaves and flowers of rape. Among varieties with various oil contents. BnLACS4 was expressed differently both in seeds and pod coats at 35 d after artificial pollination (DAP). The expression pattern suggested that BnLACS4 might be involved in the biosynthesis of lipids and oil accumulation in rapeseed.
