基因LSM2甲基化在上皮性卵巢癌发病中的作用研究

摘要 目的:检测上皮性卵巢癌患者肿瘤组织DNA中基因LSM2启动子区CpG岛(CpG island,CGI)的甲基化状态,探讨其在上皮性卵巢癌发病机制中的作用。方法:选取50例上皮性卵巢癌患者和25例卵巢良性病变患者作为实验组和对照组,提取肿瘤组织DNA,用Taqman实时荧光定量PCR(MethyLight)方法检测基因LSM2启动子区CGI在肿瘤组织DNA中的甲基化状态。结果:上皮性卵巢癌和卵巢良性病变患者肿瘤组织DNA中,基因LSM2启动子区CGI的甲基化率分别为10%(5/50)和32%(8/25),与卵巢良性病变患者相比,上皮性卵巢癌患者基因LSM2启动子区CGI的甲基化程度降低,差异具有统计学意义(χ2=5.318,P<0.05)。结论:基因LSM2启动子区CGI在上皮性卵巢癌患者肿瘤组织中呈现低甲基化改变,有可能成为新的上皮性卵巢癌特异性的低甲基化肿瘤标志物。
