摘要
目的探讨子宫内膜癌组织中COX-2表达的上调与肿瘤细胞增殖的关系。方法分析60例子宫内膜癌患者,应用蛋白印迹法分别测定子宫内膜癌组织中COX-2、VEGF的蛋白表达水平,应用流式细胞技术分析肿瘤细胞凋亡、增殖情况。其中无肌层浸润组中30例子宫内膜癌病例均为早期无肌层侵袭和淋巴转移者;肌层浸润组中30例子宫内膜癌病例均有深肌层侵袭和(或)伴有淋巴转移者。对照组入选病例均无子宫内膜病变。结果①研究组1和2中COX-2、VEGF蛋白表达水平均高于对照组,且有统计学意义(P<0.05);②研究组1和2之间COX-2、VEGF的蛋白表达水平,且有显著统计学意义(P<0.05);③研究组1和2中肿瘤组织PI指数均高于对照组,且有统计学意义。结论子宫内膜癌组织中COX-2的上调促进细胞增殖,抑制细胞凋亡,促进新生血管和淋巴管生成,从而影响子宫内膜癌肌层浸润和淋巴转移。
Objective To investigate relation between the high expressions of COX-2 and cell proliferation in the endometrial cancer. Methods A retrospective analysis of 60 endometrial cancer patients were selected as the subject group 1 and 2, and 30 normal women as control group.The expressions of COX-2 and VEGF in different endometrial tissues were examined using real time RT-PCR, western-blot. Results ①The protein levels of COX-2 and VEGF in subject group are higher than control group,while In statistical terms, significant necessarily mean important (P〈 0.05).② Between subject group 1 and 2, the protein levels of COX-2 and VEGF were significantly different .In statistical terms, significant necessarily mean important (P〈0.05).③The PI in subject group are higner than control group, while In statistical terms, significant necessarily mean important (P〈0.05). Conclusions The high expression of COX-2 in endometrial cancer tissues may upregulate the expression of VEGF, and they play an important role in inducing Angiogenesis and Lymphangiogenesis. Both of COX-2 and VEGF are associated with the myometrial invasion and lymphatic metastasis of endometrial cancer.
