摘要
目的:探讨胰岛素生长因子Ⅱ(IGF-Ⅱ)对人早孕滋养细胞的迁移和侵袭力的影响。方法:采用原代培养人早孕滋养细胞,划痕实验检测不同IGF-Ⅱ浓度(0μg/L,10.5μg/L,26.5μg/L,52.5μg/L,112.5μg/L)对滋养细胞迁移力的影响,Ma-trigel侵袭试验分析不同浓度IGF-Ⅱ对滋养细胞的侵袭力的影响。结果:IGF-Ⅱ可以明显促进原代培养的人早孕滋养细胞的迁移和侵袭力,并随着药物浓度和时间的增加而增强,各组间有显著性差异。结论:IGF-Ⅱ可明显促进人早孕滋养细胞的迁移和侵袭能力,可能在绒毛外滋养细胞的入侵和胚胎着床过程中具有重要作用。
Objective:To explore the migration and invasion of first -trimester human cytotrophoblast cells induced by recombination human insulin - like growth factor Ⅱ( IGF - Ⅱ ) . Methods : Wound healing assay was used to detect the effect of various concentration rhlGF - Ⅱ(0μg/L, 10. 5 μg/L, 26. 5 μg/L, 52.5 μg/L, 112. 5 μg/L) on the primary - cultured cytotrophoblast cells. Then, matrigel invasion assay was applied to investigate the invasion effect of various concentration rbIGF - l] on the cytotrophoblast cells. Results:rhIGF - II stimulated migration and invasion of the primary cultured cytotrophoblast in vitro, there was positive correlation between the effect and concentration, time, and there was significant difference between different concentrations. Conclusion : rhIGF - Ⅱ stimulates migration and invasion of first - trimester human cytotrophoblast cells, which plays an important roles in extravillous trophoblast invasiveness and embryo nidation.
