基因芯片检测宫颈上皮内瘤变和浸润性鳞癌中HPV基因型别 被引量:1

Detection of human papillomavirus genotypes by HPV DNA chip in cervical intraepithelial neoplasm and invasive squamous cell carcinoma

摘要 目的:探讨宫颈上皮内瘤变(CIN)和浸润性鳞状细胞癌(SCC)中HPV的基因类型。方法:采用HPV DNA基因芯片检查77例存档宫颈标本(32例CIN和45例SCC)中HPV基因类型。结果:77例宫颈标本中HPV总的阳性率为72.7%,CINⅠ、CINⅡ、CINⅢ及SCC的HPV阳性率分别为50%、60%、75%和80%,HPV感染中16型最多(73.2%),SCC中多重感染3例(6.7%)。结论:HPV感染与CIN和SCC密切相关,随着组织学分级的增加HPV阳性率呈增加的趋势,HPV-16型是主要的感染类型。基因芯片可同时检测多种HPV的基因型别,灵敏度高、特异性强,在防治宫颈上皮肿瘤中具有较大的应用价值。 Objective: To explore the human papillomavirus (HPV) genotypes in cervical intraepithelial neoplasm (CIN) and invasive squamous cell carcinoma (SCC) . Methods: The HPV genotypes were detected by HPV DNA chip in archival tissues from 77 cervical specimens, including 32 cases of CIN and 45 cases of invasive SCC. Results: The overall HPV positive rate was 72.7% in all specimens, according to the histologic grade, the HPV positive rates were 50% , 60% , 75% and 80% in CIN I , CIN Ⅱ , CIN Ⅲ and SCC, respectively. HPV-16 was the most frequent genotype (73.2%), and multiple HPV infection was detected in 3 cases (6.7%) of SCC. Conclusion: HPV infection is closely related to CIN and SCC, and the HPV - 16 was the most frequent genotype. HPV DNA chip can detect and classify different HPV genotypes with high sensitivities and speeificities simultaneously, and might be beneficial for prevention and management of cervical epithelial neoplasia.
